What is Tesamorelin?

Tesamorelin is a synthetic 44-amino acid linear peptide that is structurally identical to the full-length sequence of human growth hormone-releasing hormone (GHRH 1-44). Unlike truncated GHRH analogues that contain only the bioactive N-terminal fragment, Tesamorelin includes the complete GHRH sequence, providing the full native peptide for endocrine research.

Growth hormone-releasing hormone (GHRH), also known as somatocrinin or growth hormone-releasing factor (GRF, GHRF), is produced by neurosecretory neurons in the arcuate nucleus of the hypothalamus. It is released into the hypothalamic-pituitary portal circulation and transported to the anterior pituitary, where it binds to GHRH receptors on somatotroph cells.

Mechanism of Action

Tesamorelin binds specifically to the GHRH receptor (GHRH-R), a class B G-protein-coupled receptor expressed on the surface of somatotroph cells in the anterior pituitary. Receptor binding activates the Gs alpha subunit, stimulating adenylyl cyclase and increasing intracellular cAMP. The resulting activation of protein kinase A (PKA) and downstream transcription factors (including CREB and Pit-1) promotes the synthesis and pulsatile secretion of growth hormone (GH).

The GH released from the pituitary then travels through the systemic circulation to stimulate hepatic production of insulin-like growth factor 1 (IGF-1), which mediates many of the peripheral effects of GH signalling. This GHRH-GH-IGF-1 axis is the central endocrine pathway regulating growth, metabolism, and body composition throughout life.

Reconstitution Guidelines

For experienced laboratory personnel only. This lyophilised powder must be reconstituted with bacteriostatic water prior to research use.

  • Allow the vial to equilibrate to room temperature (15–25°C) before reconstitution.
  • Wipe the rubber stopper with an alcohol swab and allow it to dry.
  • Using a sterile syringe, add the required volume of Bacteriostatic Water by injecting slowly down the inner wall of the vial — do not aim the stream directly at the lyophilised cake.
  • Gently swirl the vial until the powder is fully dissolved. Do not shake vigorously, as this can denature the peptide.
  • Visually inspect the reconstituted solution — it should be clear and free of particulate matter.
  • Store reconstituted solutions at 2–8°C and use within the timeframe defined in your research protocol. Discard any unused solution after the recommended period.
