What is SemaG?

SemaG is a 31-amino acid synthetic analogue of the naturally occurring incretin hormone GLP-1 (glucagon-like peptide-1). It shares approximately 94% sequence homology with native human GLP-1 but incorporates two key structural modifications: a substitution of alanine to alpha-aminoisobutyric acid at position 8 (protecting against DPP-4 enzyme degradation), and the attachment of a C18 fatty diacid side chain via a glutamic acid spacer at lysine 26 (enabling albumin binding for prolonged half-life). These modifications make SemaG one of the most extensively characterised GLP-1 receptor agonists in the metabolic research literature.

With a molecular weight of approximately 4,113 Da, SemaG binds specifically to the GLP-1 receptor (GLP-1R), a class B G-protein-coupled receptor expressed in pancreatic islets, the central nervous system, the gastrointestinal tract, and various peripheral tissues.

Mechanism of Action

SemaG functions as a full agonist at the GLP-1 receptor. Receptor binding triggers a conformational change that activates the Gs alpha subunit, stimulating adenylyl cyclase and increasing intracellular cyclic AMP (cAMP) levels. This cAMP signalling cascade activates downstream effectors including protein kinase A (PKA) and exchange protein activated by cAMP (EPAC). The resulting cellular effects — which vary by tissue type — are the subject of ongoing research across multiple disciplines.

The fatty acid side chain enables non-covalent binding to serum albumin, which significantly slows renal clearance and maintains elevated peptide levels for extended periods in research models — a key advantage over shorter-acting GLP-1 analogues.

Product Description

This 6mg vial contains lyophilised SemaG peptide as a sterile white to off-white powder or cake. Each vial is individually sealed under vacuum in a glass container with rubber stopper and aluminium crimp seal. The 6mg quantity is suitable for multiple research applications or protocols requiring precise dosing.

The lyophilised formulation provides stability during storage. Reconstitution with an appropriate sterile diluent is required prior to use in laboratory protocols.

Handling Guidelines
