What is SemaG?

SemaG is a 31-amino acid synthetic analogue of the naturally occurring incretin hormone GLP-1 (glucagon-like peptide-1). It shares approximately 94% sequence homology with native human GLP-1 but incorporates two key structural modifications: a substitution of alanine to alpha-aminoisobutyric acid at position 8 (protecting against DPP-4 enzyme degradation), and the attachment of a C18 fatty diacid side chain via a glutamic acid spacer at lysine 26 (enabling albumin binding for prolonged half-life). These modifications make SemaG one of the most extensively characterised GLP-1 receptor agonists in the metabolic research literature.

With a molecular weight of approximately 4,113 Da, SemaG binds specifically to the GLP-1 receptor (GLP-1R), a class B G-protein-coupled receptor expressed in pancreatic islets, the central nervous system, the gastrointestinal tract, and various peripheral tissues.

Mechanism of Action

SemaG functions as a full agonist at the GLP-1 receptor.
